Complaint is a general word and can refer to serious or minor problems, practical problems or personal unhappiness about sth. A complaint can be informal, and made to a friend, a colleague, etc. or it can be filed or lodged more formally, often in writing, to a company or organization. A grievance is a strong feeling that sth is unfair or that you have been treated unfairly. It is often a feeling which has developed over a period of time.
